Specific Seahorse XF Assays Using Permeabilized Cells and Isolated Mitochondria
Cellular Bioenergetics lie at the epicenter that drive cell function in health and disease. Agilent Seahorse XF Analyzers have been the cornerstone of metabolic studies and have been widely used with whole cells, tissues, spheroids, permeabilized fibers, and isolated mitochondria. There are ready-to-use and gold standard XF kits and assays that enable the users to examine metabolic phenotypes and changes in the cells. In this session, the focus will be on elaboration of techniques for detailed characterization of key components in mitochondrial function including transporters, enzymes, and electron transport chain complexes requires analyzing isolated mitochondria or detergent permeabilized cells in a single chamber using an oxygen electrode.
In this talk you will learn:
- An introduction and discussion on permeabilizing cells with the Agilent XF Plasma Membrane Permeabilization (PMP) reagent. PMP can be used with adherent cells and allows one to do more varied experiments with genetically modified cell lines.
- A user presentation who will discuss about the applications of PMP in assessing mitochondrial bioenergetic function in intact and permeabilized primary cells, cell lines, and blood cells.
- A brief discussion on isolating high-quality mitochondria from tissues and the assays one can use, using much less mitochondrial mass for the same amount of data than other methods. Various applications of using isolated mitochondria will also be discussed.
